It seems that the flowers that Donald has, called Magnolia Ridens in the first caption and "atomic flower" in the second one, make him laugh. So when Donna Filomena asks him about her hat, Donald start laughing for the flower and she thinks he is making fun of her. Then she hits him with the umbrella until it breaks. And then the policeman arrives, and both him and her become affected by the flower effect.

she only appears in a few panels for a gag, Donald salutes her as a friend, I guess she might just be Clara Cluck.

Since Donald refers to her as Donna/Mrs., which as I understand is used with older/married people, and her can arguably be translated to Philomena Cluck, could this character be intended as Clara's mother?
I guess the use of the word "donna" here is more a kind of reverence/esteem, since she is presented in the panel as a "noble lady". And that plays a nice contrast with the reaction she is going to have in the later panels.

And yes, the early Disney stories were really weird. This story is a good example of some of the total randomness that was going on in the early '50. Someone has stolen uncle Scrooge castle, made with all kind of precious materials, and has left a message. From that Mickey understand that the castle is now in the Valley of the Enchantment (Valle dell'Incanto). With Donald and Pluto they start they journey, but are then attacked by pirate Pete. They are able to escape on an air baloon, and reach the valley between high mountains, where they find a masked Pete, which is the king of the valley. And this is only the first of the three episodes.
The thing that differentiate this from other early stories is that this is the first appearence of Scrooge in an Italian story, only 5 years after his creation, and he is there only to start the plot.
